چکیده

ATP and ADP inactivate rabbit liver fructose 1,6-diphosphatase and change the nature of inhibition by AMP from sigmoidal to hyperbolic, with a concomitant increase in sensitivity to the inhibitor. 3-Phosphoglycerate both prevents and reverses the inactivation and restores the sigmoidal inhibition characteristics. The above kinetic behavior, which was observed with physiological concentrations of liver metabolites, together with the known upward flux in 3-phosphoglycerate concentration during gluconeogenesis may provide a possible physiological mechanism for regulation of liver fructose 1,6-diphosphatase activity.

Role of enzyme-enzyme interactions in the regulation of glycolysis. Inactivation of D-fructos 1,6-diphosphatase by kidney cortex mitochondria.

Particulate preparations obtained from a number of tissues, including liver, kidney, and muscle inactivated renal Dfructose 1,6-diphosphatase when they were incubated with the enzyme in the presence of adenosine triphosphate, magnesium ion, and cysteine. Purified swine kidney Dfructose 1,6diphosphatase and rat kidney uridine diphosphate-D-glucose-glycogen glucosyltransferase were inactivated by...

Kinetic properties of rat liver pyruvate kinase at cellular concentrations of enzyme, substrates and modifiers.

Kinetic properties of rat liver pyruvate kinase type I at pH7.5 and 6.5 were studied with physiological ranges of substrates, modifiers and Mg(2+) concentrations at increasing enzyme concentrations, including the estimated cellular concentrations (approx. 0.1mg/ml). Enzyme properties appear unaffected by increased enzyme concentration if phosphoenolpyruvate, fructose 1,6-diphosphate and inhibit...

Properties of Phosphofructokinase from Rat Liver and Their Relation to the Control of Glycolysis and Gluconeogenesis.

1. Phosphofructokinase from rat liver has been partially purified by ammonium sulphate precipitation so as to remove enzymes that interfere in one assay for phosphofructokinase. The properties of this enzyme were found to be similar to those of the same enzyme from other tissues (e.g. cardiac muscle, skeletal muscle and brain) that were previously investigated by other workers. 2. Low concentra...

Fish liver fructose 1,6-diphosphatase. 3. The effect of temperature on the regulatory properties of the carp (Cyprinus carpio) liver enzyme.

Fructose 1,6-diphosphatase has been partially purified from the liver of the carp, Cyprinus carpio. The enzyme shows the general characteristics of the native form of most fructose 1,6-diphosphatases: maximal activity at neutral pH ; high degree of specificity for its substrate, fructose 1,6-diphosphate; requirement for a divalent cation (Mg2+ or Mn2+); activation by monovalent cations (K+ or N...

Temperature and the regulation of enzyme activity in poikilotherms. Regulatory properties of fructose diphosphatase from muscle of the Alaskan king-crab.

1. The properties of fructose diphosphatase from skeletal muscle of the Alaskan king-crab (Paralithodes camtschatica) were examined over the physiological temperature range of the animal. 2. King-crab muscle fructose diphosphatase is first activated by Na(+) and NH(4) (+) and is then partially inhibited by these cations at concentrations higher than 10mm at 0 degrees , 8 degrees and 15 degrees ...
